I am now working totally in SharePoint. 30,000 users and it doesn't do everything a database does. Not much else to say. We do however use lists way over 2,000 items but then I  have two great .NET programmers here who do most of that.

You can however, use the BDC in MOSS 2007 to expose relational data etc and can buy in some useful third party products that will work out ok.

 SharePoint 2010 does however offer a huge range of increased features.

The biggest mistake you can make is to assume  that SharePoint is the answer to every problem.
